چکیده

Objective: DLBS3233 is a standardized extract combination containing Lagerstroemia speciosa and Cinnamomum burmannii. The effect of DLBS3233 on adipocyte differentiation was examined in this study. Methods: 3T3-L1 pre-adipocyte was used to investigate gene expression using the real-time reverse transcription polymerase chain reaction (RTPCR) method. Oil red-O staining for detecting lipid formation was also carried out in this experiment. Results: DLBS3233 caused cell differentiation of 3T3-L1 pre-adipocytes into adipocytes which were indicated by positive results on staining cells with oil red-O on day 6 of the differentiation process. Analysis of gene expressions associated with adipogenesis (C/EBP-α, PPAR-γ, C/EBP-δ, FASn and adiponectin) showed an increase compared to control. In this study, DLBS3233 at a concentration of 5 μg/ml exhibited better differentiation effect than DLBS3233 at a concentration of 10 μg/ml. Conclusion: DLBS3233 can stimulate differentiation of 3T3-L1 pre-adipocytes into adipocytes.

Fifty Percent Ethanolic Extract of Momordica charantia Inhibits Adipogenesis and Promotes Adipolysis in 3T3-L1 Pre-Adipocyte Cells

Background: Natural products have gained importance recently for the treatment of obesity and its complications, partly because of the side effects of modern drugs.Hence, we aimed to study and compare the effect of varying concentrations of Momordicacharantiaon adipogenesis and adipolysis using 3T3-L1 pre-adipocyte cell lines. Methods: 3T3-L1 pre-adipocytes were procured from the National Ce...

Metformin and Pioglitazone Reduce Gene Expression of Inflammatory Factors in Insulin Resistant and Hypertrophied Adipocytes

Objective: In obesity, chronic low grade inflammation, created by induction of pro-inflammatory markers, causes adipocyte dysfunction in adipose tissue. Adipocytes dysfunction is associated with various diseases including insulin resistance and obesity. In obesity, inflammatory factors such as osteopontin (OPN), angiopoietin-like protein 2 (Angptl2) and transforming growth factor-β (TGF-β) are...

Ginsenoside Rg5: Rk1 Exerts an Anti-obesity Effect on 3T3-L1 Cell Line by the Downregulation of PPARγ and CEBPα

Background: Obesity, a global health problem and a chronic disease, is associated with increased risk of developing type 2 diabetes and coronary heart diseases. A wide variety of natural remedies have been explored for their obesity treatment potential. Objective: The anti-adipogenic effect of ginsenoside Rg5:Rk1 (Rg5:Rk1) on 3T3-L1 mature adipocytes was investigated. Materials and ...

Inhibitory effect of microRNA-24 on fatty acid-binding protein expression on 3T3-L1 adipocyte differentiation.

We examined the effect of microRNAs on 3T3-L1 adipocyte differentiation and expression of adipocyte-specific gene fatty acid-binding protein 4 (FABP4). We screened and identified adipo-related microRNAs during 3T3-L1 adipocyte differentiation with a microRNA microarray. High expression plasmids of miR-24 and miR-21 were constructed and transfected into 3T3-L1 preadipocytes by lipofectamine. The...

Development of a Novel Anti-Obesity Compound with Inhibiting Properties on the Lipid Accumulation in 3T3-L1 Adipocytes

Background: Obesity as a developing global challenge can be characterized by increase in adipocyte number and size arising from adipogenesis. Control of adipogenesis, as a potential strategy, can prevent and manage obesity. So far, the effectiveness of herbal medicine and active ingredients therapies for obesity and metabolic syndrome treatment has been investigated. In this study, a novel comb...
